Although the potential of hPSCs is truly outstanding, fulfilling this potential depends on our ability to efficiently generate functional cell types from them. One of the key problems in the stem cell directed differentiation field is that we are able to efficiently generate alveolar epithelial type II (AT2) cells but cannot efficiently generate alveolar epithelial type I (AT1) cells. On the contrary, it is nearly impossible to keep primary AT2 cells transdifferentiating to AT1 cells after isolating and culturing the cells. We have recently discovered a factor that can delay transdifferentiation from AT2 cells to AT1 cells. By identifying and revealing the mechanism of the factor, we will potentially have a tool to specifically generate AT1 or AT2 cells.

[/et_pb_text][/et_pb_column][/et_pb_row][et_pb_row column_structure=”1_2,1_2″ use_custom_gutter=”on” gutter_width=”0″ make_equal=”on” disabled_on=”on|on|off” admin_label=”Stem Cell therapy-Desktop” module_class=” et_pb_row_fullwidth” _builder_version=”4.9.0″ background_color=”#000000″ z_index=”126″ width=”100%” width_tablet=”100%” width_phone=”” width_last_edited=”on|desktop” max_width=”100%” max_width_tablet=”100%” max_width_phone=”” max_width_last_edited=”on|desktop” custom_margin=”-50px|auto|0px|auto|false|false” custom_padding=”0px||22px|||” make_fullwidth=”on” locked=”off”][et_pb_column type=”1_2″ _builder_version=”3.25″ custom_padding=”5%|5%|5%|5%” custom_padding__hover=”|||”][et_pb_text admin_label=”Title” _builder_version=”4.9.0″ text_font=”||||||||” header_font=”||||||||” header_2_font=”Poppins|600|||||||” header_2_font_size=”36px” header_2_line_height=”1.2em” background_layout=”dark” max_width=”750px” custom_margin=”||||false|false” animation_style=”fade” animation_direction=”left” animation_starting_opacity=”100%”]

Stem Cell Therapy

[/et_pb_text][et_pb_divider color=”#fdb467″ divider_position=”center” divider_weight=”3px” _builder_version=”3.2″ max_width=”50px” animation_style=”slide” animation_intensity_slide=”10%” animation_starting_opacity=”100%”][/et_pb_divider][et_pb_text _builder_version=”4.9.0″ text_font=”Poppins||||||||” text_text_color=”rgba(255,255,255,0.7)” text_font_size=”18px” text_line_height=”1.8em” max_width=”750px” custom_margin=”10px|||||” custom_padding=”|0px|1px|||” animation_style=”fade” animation_direction=”left” animation_starting_opacity=”100%”]

In the field of regenerative medicine, one of the biggest challenges is regenerating organs comparable to the originals that form during embryonic development. In our lab, we are exploring the potential of using lung organoids as the regenerative bud for lung regeneration.

[/et_pb_text][/et_pb_column][et_pb_column type=”1_2″ _builder_version=”3.25″ custom_padding=”|||” custom_padding__hover=”|||”][/et_pb_column][/et_pb_row][et_pb_row column_structure=”1_2,1_2″ use_custom_gutter=”on” gutter_width=”0″ make_equal=”on” admin_label=”Trachea bioengineering” module_class=” et_pb_row_fullwidth” _builder_version=”4.9.0″ background_color=”#1e1d1c” z_index=”199″ width=”100%” width_tablet=”100%” width_phone=”” width_last_edited=”on|desktop” max_width=”100%” max_width_tablet=”100%” max_width_phone=”” max_width_last_edited=”on|desktop” custom_margin=”-23px|0px|0px|0px|false|false” custom_padding=”0px|0px|0px|0px|false|false” make_fullwidth=”on”][et_pb_column type=”1_2″ _builder_version=”3.25″ custom_padding=”|||” custom_padding__hover=”|||”][et_pb_slider show_arrows=”off” _builder_version=”4.9.0″ _module_preset=”default” background_color=”#1e1d1c” min_height=”800px” custom_margin=”||||false|false” auto=”on” auto_speed=”3000″ border_width_all=”5px” border_color_all=”#1e1d1c”][et_pb_slide _builder_version=”4.9.0″ _module_preset=”default” background_color=”#000000″ background_enable_color=”on” background_image=”https://labs.icahn.mssm.edu/yawen-chen-lab/wp-content/uploads/sites/438/2022/07/Picture37.jpg” background_enable_image=”on” background_size=”contain” sticky_transition=”on”][/et_pb_slide][et_pb_slide _builder_version=”4.9.0″ _module_preset=”default” background_color=”#000000″ background_enable_color=”on” background_image=”https://labs.icahn.mssm.edu/yawen-chen-lab/wp-content/uploads/sites/438/2022/07/Picture38.jpg” background_enable_image=”on” background_size=”contain” sticky_transition=”on”][/et_pb_slide][/et_pb_slider][/et_pb_column][et_pb_column type=”1_2″ _builder_version=”3.25″ custom_padding=”5%|5%|5%|5%” custom_padding__hover=”|||”][et_pb_text admin_label=”Title” _builder_version=”4.9.0″ text_font=”||||||||” header_font=”||||||||” header_2_font=”Poppins|600|||||||” header_2_font_size=”36px” header_2_line_height=”1.2em” background_layout=”dark” max_width=”750px” custom_margin=”||||false|false” animation_style=”fade” animation_direction=”left” animation_starting_opacity=”100%”]

Trachea Transplantation

[/et_pb_text][et_pb_divider color=”#fdb467″ divider_position=”center” divider_weight=”3px” _builder_version=”3.2″ max_width=”50px” animation_style=”slide” animation_intensity_slide=”10%” animation_starting_opacity=”100%”][/et_pb_divider][et_pb_text _builder_version=”4.9.0″ text_font=”Poppins||||||||” text_text_color=”rgba(255,255,255,0.7)” text_font_size=”18px” text_line_height=”1.8em” max_width=”750px” custom_margin=”15px|||||” custom_padding=”0px||0px||false|false” animation_style=”fade” animation_direction=”left” animation_starting_opacity=”100%”]

With few definitive treatment options available, tracheal transplantation is widely proposed for patients suffering end-stage upper airway diseases. However successful tracheal transplantation surgery remains a major challenge in the field of thoracic surgery and regenerative medicine. Previous trials in patients had shown that recellularization (re-cell) of the tracheal scaffold is crucial to obtain functional long-segment tracheal grafts. Grafts without substantial epithelial cell coverage uniformly fail to remodel appropriately and lead to graft failure. One of the biggest hurdles to successful creation of tissue-engineered tracheas is an inability to isolate and expand a sufficient number of epithelial cells from patients for re-cell. For the past two years, we have developed a method that enables us to expand basal cells, the stem cell of the airways, in vitro from human pluripotent stem cells. The immediate application is to serve as an unlimited source of cells for tissue-engineered tracheal/airway reconstruction. We are evaluating the potential of the stem cell-generated basal cells to serve as an unlimited epithelial cell source for tracheal/airway reconstruction and to create a bioengineered tracheal graft with functional epithelium.

[/et_pb_text][/et_pb_column][/et_pb_row][et_pb_row column_structure=”1_2,1_2″ use_custom_gutter=”on” gutter_width=”0″ make_equal=”on” disabled_on=”on|on|off” admin_label=”Injury model-Desktop” module_class=” et_pb_row_fullwidth” _builder_version=”4.9.0″ background_color=”#000000″ width=”100%” width_tablet=”100%” width_phone=”” width_last_edited=”on|desktop” max_width=”100%” max_width_tablet=”100%” max_width_phone=”” max_width_last_edited=”on|desktop” custom_margin=”-4px|auto|0px|auto|false|false” custom_padding=”0px||22px|||” make_fullwidth=”on”][et_pb_column type=”1_2″ _builder_version=”3.25″ custom_padding=”5%|5%|5%|5%” custom_padding__hover=”|||”][et_pb_text admin_label=”Title” _builder_version=”4.9.0″ text_font=”||||||||” header_font=”||||||||” header_2_font=”Poppins|600|||||||” header_2_font_size=”36px” header_2_line_height=”1.2em” background_layout=”dark” max_width=”750px” custom_margin=”||||false|false” animation_style=”fade” animation_direction=”left” animation_starting_opacity=”100%”]

Injury models

[/et_pb_text][et_pb_divider color=”#fdb467″ divider_position=”center” divider_weight=”3px” _builder_version=”3.2″ max_width=”50px” animation_style=”slide” animation_intensity_slide=”10%” animation_starting_opacity=”100%”][/et_pb_divider][et_pb_text _builder_version=”4.9.0″ text_font=”Poppins||||||||” text_text_color=”rgba(255,255,255,0.7)” text_font_size=”18px” text_line_height=”1.8em” max_width=”750px” custom_margin=”10px|||||” custom_padding=”||1px|||” animation_style=”fade” animation_direction=”left” animation_starting_opacity=”100%”]

To study the mechanism of injury and repair in the distal lung, we have created injury models using lung organoids.
